AICAR (Acadesine) is an adenosine analogue and an AMPK activator. AICAR regulates glucose and lipid metabolism and inhibits proinflammatory cytokine and iNOS production. AICAR is also an inhibitor of autophagy, YAP and mitophagy.AICAR, the full name is 5-aminoimidazole-4-carboxamide1-β-d-ribofuranoside, also known as AICARiboside or Acadesine is an activator of AMP-activatedproteinkinase(AMPK) that permeates cell membrane.AMPK is a key protein in metabolic regulation. When the energy supply is insufficient, the AMP/ATP ratio is upregulated, and AMPK will be activated to inhibit anabolism. AICAR can activate AMPK without affecting ATP, ADP and AMP levels. At the cellular or animal level, AICAR promotes non-insulin-dependent glucose intake in skeletal muscle by activating AMPK. Aicar-induced glucose intake in skeletal muscle was not blocked by inhibitors of PI3K.
